Biological Background: Glycine dehydrogenase (decarboxylating), mitochondrial Function and Localization
- The protein is also known as Glycine cleavage system P protein, Glycine decarboxylase, or Glycine dehydrogenase (aminomethyl-transferring), encoded by the GLDC gene (also designated GCSP).
- It catalyzes the degradation of glycine by binding the alpha-amino group of glycine through its pyridoxal phosphate cofactor; CO₂ is released and the remaining methylamine moiety is transferred to the lipoamide cofactor of the H protein (GCSH).
- GLDC is a key component of the glycine cleavage system, which is essential for the metabolism of glycine, a major amino acid involved in various metabolic pathways.
- It is localized in the mitochondrion, indicating its role in mitochondrial energy metabolism.
- The protein belongs to the class of oxidoreductases and requires pyridoxal phosphate as a cofactor.
- Post-translational modifications such as acetylation have been identified through proteomics.
- Disease-associated variants have been reported for GLDC.
- The molecular weight of the mature protein is approximately 113 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 580–749, a C-terminal fragment; the antibody may recognize epitopes within this region.
- For Western blotting, a band around 113 kDa is expected; consider appropriate gel percentages and transfer conditions for high molecular weight proteins.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), mitochondrial localization may result in a punctate cytoplasmic staining pattern; validate in the relevant sample system.
- The antibody may be suitable for ELISA-based assays; users should optimize coating and detection conditions.
