Biological Background: GK2 Function and Localization

• GK2 (Glycerol kinase 2) is a testis-specific member of the glycerol kinase family that catalyzes ATP-dependent phosphorylation of glycerol to glycerol-3-phosphate.
• It plays a crucial role in glycerol uptake and metabolism, and is essential for male fertility by participating in spermatogenesis and mitochondrial sheath formation.
• Required for proper organization of mitochondria into the crescent-like mitochondrial sheath during sperm development.
• Can induce mitochondrial clustering through interactions with PLD6 and up-regulation of phosphatidic acid synthesis in the mitochondria. Related references: PMID:28852571
• Subcellularly localized to the mitochondrial outer membrane and cytoplasm.
• Testis-specific in expression, with protein detected in the midpiece of spermatozoa.

Experimental Guidance and Technical Tips

• The immunogen corresponds to amino acids 1–150 of human GK2; thus, the antibody may detect both full-length GK2 and any N-terminal variants.
• In Western blotting, a band at ~61 kDa is expected; it is advisable to use testis or sperm protein extracts as positive controls to confirm specificity.
• For immunofluorescence/ICC, permeabilization is required to access the cytoplasmic and mitochondrial outer membrane pools of GK2; testis tissue sections or sperm smears are recommended.
• ELISA assays may be used for quantification or screening; cross-reactivity with mouse and rat GK2 should be verified if using these species' samples.
