Biological Background: GITR/TNFRSF18 Function and Localization
- GITR (TNFRSF18) is a member of the tumor necrosis factor receptor superfamily, also known as AITR or CD357.
- It functions as a receptor for TNFSF18 (GITR ligand) and is implicated in T-cell-mediated immune responses.
- Involved in interactions between activated T-lymphocytes and endothelial cells, and in regulating T-cell receptor-mediated cell death.
- Mediates NF-kappa-B activation through the TRAF2/NIK signaling pathway.
- Localized to the cell membrane, but a secreted isoform may also exist, as indicated by alternative splicing.
- Expressed predominantly in lymph node, peripheral blood leukocytes, and weakly in spleen.
- Contains characteristic TNF receptor features including disulfide bond-rich repeats and a transmembrane helix; is a glycoprotein.
Experimental Guidance and Technical Tips
- The immunogen covers the extracellular domain (aa 26-255), which may facilitate recognition of both membrane-bound and soluble forms.
- For Western blotting, consider using lysates from lymph node or leukocyte cell lines where GITR expression is reported.
- Given the reported mouse cross-reactivity, validate antibody specificity in the relevant species and sample system before critical experiments.
- For ELISA, consider an indirect format with recombinant GITR protein as the capture target.
