Biological Background: Tumor necrosis factor ligand superfamily member 18 Function and Localization
- Member of the tumor necrosis factor (TNF) ligand superfamily, also known as GITR ligand (GITRL), AITRL, or TL6.
- Functions as a cytokine that binds to the receptor TNFRSF18 (GITR), acting as a costimulatory molecule to lower the threshold for T-cell activation and promote T-cell proliferation.
- Plays a key role in mediating interactions between activated T lymphocytes and endothelial cells, enhancing leukocyte adhesion through upregulation of VCAM1 and ICAM1. Related references: PMID:23892569
- Mediates activation of NF-kappa-B signaling and triggers increased phosphorylation of STAT1. Related references: PMID:23892569
- Regulates monocyte migration from the splenic reservoir to sites of inflammation.
- Localizes to the cell membrane as a single-pass type II membrane protein.
- Highly expressed in the small intestine, ovary, testis, kidney, and endothelial cells.
- Contains a disulfide bond and is subject to glycosylation; predicted molecular weight of approximately 20.3 kDa.
Experimental Guidance and Technical Tips
- For Western blotting, a band around 20–25 kDa is anticipated, considering potential glycosylation. Validate the antibody in your relevant sample system (human, mouse, or rat) and optimize blocking/detection conditions.
- For ELISA, consider using the recombinant immunogen (aa 52–177) as a coating antigen or pair with a matched detection antibody. Establish optimal dilutions for both capture and detection steps.
- As a polyclonal antibody, it may exhibit batch-to-batch variation; we recommend titrating the antibody for your specific application and including appropriate positive and negative controls.
