Biological Background: GIT2 Function and Localization
- GIT2, also known as CAT-2, CAT2, PKL, or GRK-interacting protein 2, is a member of the GIT family of ARF GTPase-activating proteins.
- Functions as a GTPase-activating protein (GAP) for ADP-ribosylation factor (ARF) proteins, including ARF1, negatively regulating their activity.
- Acts as a scaffold linking G protein-coupled receptor kinases (GRKs) and p21-activated kinases (PAKs) to focal adhesion complexes.
- Contains ankyrin repeats and a coiled-coil domain that mediate protein-protein interactions, as well as a zinc-finger motif.
- Alternative splicing produces multiple isoforms; annotated as a phosphoprotein in several proteomics studies.
- Plays roles in cell adhesion, migration, and receptor internalization through modulation of ARF-dependent membrane trafficking.
