Biological Background: GIMAP5 Function and Localization
- GIMAP5 (GTPase IMAP family member 5) is a member of the immune-associated nucleotide-binding protein (IAN) family, also known as IAN-5, hIAN5, and IMAP3. It is a GTP-binding protein with a transmembrane domain.
- Plays a critical role in T lymphocyte development, particularly in the optimal generation of CD4/CD8 double-positive thymocytes, as suggested by similarity data.
- Acts as an inhibitor of glycogen synthase kinase-3 alpha (GSK3A) by sequestering GSK3A in cytoplasmic vesicles, which impairs its nuclear translocation and downstream transcriptional programs, thereby regulating the DNA damage response during T cell proliferation. - Required for the survival of peripheral T cells, natural killer (NK) cells, and NKT-cell development, and is essential for the maintenance of normal liver function, based on similarity evidence.
- May promote the survival of mature T lymphocytes upon cytokine withdrawal and regulate calcium homeostasis by modulating lysosomal Ca²⁺ stores.
- Localizes to the membranes of lysosomes, endosomes, and multivesicular bodies, indicating a role in intracellular membrane trafficking.
- Widely expressed with high levels in lymph node and spleen; highly expressed in T lymphocytes (CD4⁺ and CD8⁺ T-cells) and monocytes, but very low in B-lymphocytes. Related references: PMID:14724691
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 190-270 of human GIMAP5; this region may be conserved across splicing variants. For WB, validate in the relevant cell type or tissue known to express GIMAP5.
- For western blotting, a band around 35 kDa is expected in human, mouse, and rat samples. Consider using a positive control such as Jurkat T-cell lysate.
- In ELISA, assess coating antigen concentrations and detection antibody dilutions for optimal performance; the antibody may be used for capture or detection in sandwich ELISAs targeting GIMAP5.
- Since the antibody is polyclonal, it may recognize multiple epitopes; confirm specificity via knockdown or overexpression experiments if needed.
