Biological Background: GIGYF2 Function and Localization
- GIGYF2 (GRB10-interacting GYF protein 2), also known as PERQ2, TNRC15, and PARK11, is a 150-kDa scaffolding protein that serves as a key component of the 4EHP-GYF2 translation repressor complex.
- Within the 4EHP-GYF2 complex, GIGYF2 bridges the cap-binding protein EIF4E2 to the mRNA decay factor ZFP36/TTP, thereby linking translational repression to AU-rich element-mediated mRNA decay. Related references: PMID:22751931 | PMID:31439631 | PMID:35878012
- GIGYF2 also recruits the decapping activator DDX6 to the complex, an activity required for ZFP36/TTP-dependent downregulation of target mRNAs. Related references: PMID:31439631
- In cooperation with GRB10, GIGYF2 may modulate signaling downstream of tyrosine kinase receptors, including the insulin and IGF1 receptors. Related references: PMID:12771153
- During ribosome-associated quality control (RQC), GIGYF2 collaborates with EIF4E2 to sequester the mRNA cap and block translation initiation on stalled ribosomes, operating in parallel to the nascent chain degradation pathway. Related references: PMID:32726578
- Upon SARS-CoV-2 infection, viral non-structural protein 2 (nsp2) enhances GIGYF2–EIF4E2 binding, reinforcing translational repression of antiviral innate immune genes such as IFNB1.
- Genetic variations in GIGYF2 are associated with familial Parkinson's disease (PARK11 locus) and related neurodegenerative disorders.
- The protein undergoes multiple post-translational modifications, including phosphorylation, acetylation, ubiquitination, and methylation, which may regulate its activity and interactions.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 350-500 of human GIGYF2; consider epitope accessibility when designing verification experiments.
- For Western blot, the predicted molecular weight is ~150 kDa; optimize gel percentage and transfer conditions for high-molecular-weight proteins.
- Validate antibody reactivity in the relevant species (human, mouse, rat) and sample type (e.g., cell lysates, tissue homogenates) before primary experiments.
