Biological Background: GIF (Intrinsic Factor) Function and Localization
- GIF encodes the cobalamin binding intrinsic factor, also known as gastric intrinsic factor (IF) or INF. It is a secreted glycoprotein with a molecular weight of approximately 45 kDa (UniProt: 45,416 Da).
- Its primary function is to promote absorption of vitamin B12 (cobalamin) in the ileum by forming a complex with cobalamin and interacting with the cubilin receptor (CUBN) for receptor-mediated endocytosis.
- Expression is highly specific to the gastric mucosa, where it is secreted by parietal cells.
- The protein contains features such as a signal peptide, disulfide bonds, and glycosylation sites, and undergoes alternative splicing and phosphorylation (UniProt keywords).
- Belongs to the cobalamin transport family and is involved in cobalt ion transport. Mutations in GIF are associated with congenital intrinsic factor deficiency, a rare form of megaloblastic anemia.
- Three-dimensional structural studies have been reported (UniProt keyword: 3D-structure).
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 20–300 of human GIF, covering the N-terminal region including the signal peptide cleavage site and the cobalamin binding domain. This region is relatively conserved, supporting cross-reactivity with mouse (as indicated).
- For Western blot (WB), a band around 45 kDa is expected in gastric tissue lysates or cell lines expressing GIF. Validate the antibody in the appropriate sample system, as secretion may lead to low intracellular levels.
- For immunofluorescence/immunocytochemistry (IF/ICC), consider permeabilization to detect intracellular protein pools during synthesis. For secreted protein detection, inclusion of a secretion inhibitor (e.g., brefeldin A) may enhance the signal.
- In ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; however, pairing with a monoclonal antibody may improve specificity in sandwich ELISA formats.
- Always include appropriate positive (gastric mucosa extract) and negative (non-expressing tissue) controls, and validate the antibody’s performance in your experimental context.
