Biological Background: GGT5 (Glutathione hydrolase 5 proenzyme) Function and Localization
- GGT5 encodes the glutathione hydrolase 5 proenzyme, also known as gamma-glutamyltransferase 5 (GGT5), GGT-rel, and leukotriene-C4 hydrolase; it belongs to the gamma-glutamyltransferase family and undergoes alternative splicing, yielding multiple isoforms.
- The enzyme cleaves the gamma-glutamyl bond of extracellular glutathione tripeptide and certain glutathione-S-conjugates, releasing L-glutamate and cysteinyl-glycine. - GGT5 specifically metabolizes leukotriene C4 (LTC4) to leukotriene D4 (LTD4) during acute inflammatory responses, and also hydrolyzes S-geranylgeranyl-glutathione (GGG), acting as a negative regulator of GGG bioactivity. Together with ABCC1, it establishes GGG gradients that guide P2RY8-positive lymphocyte positioning within lymphoid follicles (By similarity).
