Biological Background: GGPS1 Function and Localization

  • GGPS1 encodes geranylgeranyl pyrophosphate synthase, also known as (2E,6E)-farnesyl diphosphate synthase, dimethylallyltranstransferase, farnesyl diphosphate synthase, farnesyltranstransferase, geranylgeranyl diphosphate synthase, or geranyltranstransferase, and is also referred to by aliases GGPPS, MDHLO, and GGPPS1.
  • This enzyme catalyzes the trans-addition of three molecules of isopentenyl pyrophosphate (IPP) onto dimethylallyl pyrophosphate (DMAPP) to form geranylgeranyl pyrophosphate, a key precursor in the biosynthesis of carotenoids and geranylated proteins.
  • Geranylgeranyl pyrophosphate synthase belongs to the FPP/GGPP synthase family of prenyltransferases.
  • Subcellular localization studies reveal that GGPS1 localizes to the cytoplasm, with enrichment in the perinuclear region and association with myofibrils, specifically the sarcomere Z line.
  • GGPS1 is abundantly expressed in testis, with lower levels observed in other tissues; it is also detected in dermal fibroblasts and skeletal muscle. Related references: PMID:10026212 PMID:9741684 PMID:32403198
  • The protein is subject to acetylation and is characterized by keywords such as metal-binding and magnesium cofactor binding.
  • Alternative splicing of GGPS1 yields multiple transcript variants, and mutations in this gene are associated with congenital muscular dystrophy and deafness.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to amino acids 164-300 of human GGPS1 (based on NP_001032354.1), covering a central region of the protein. This region may allow recognition of endogenous GGPS1 across species, but cross-reactivity should be validated in the relevant experimental system.
  • For Western blot (WB), a band around 35 kDa is expected based on the reported protein weight. However, post-translational modifications or alternative splicing may result in additional bands; validation with proper controls (e.g., positive lysates from testis or skeletal muscle) is recommended.
  • When using this antibody in ELISA, consider optimizing coating conditions and detection parameters for each specific assay format, as indirect ELISA performance can vary with different antigen presentation.
  • For tissue-specific studies, note that GGPS1 is highly expressed in testis and also found in skeletal muscle and dermal fibroblasts; expression levels should be confirmed in your cell or tissue model.
