Biological Background: GFRA1 Function and Localization
- GFRA1 (GDNF family receptor alpha-1) is a GPI-anchored coreceptor for the neurotrophic factor GDNF, essential for the development and maintenance of dopaminergic neurons. Related references: PMID:10829012 PMID:31535977
- Binding of GDNF to GFRA1 leads to autophosphorylation and activation of the RET receptor tyrosine kinase, propagating downstream signaling pathways. Related references: PMID:31535977
- The protein localizes to the cell membrane, Golgi apparatus, trans-Golgi network, endosomes, and multivesicular bodies, indicative of dynamic trafficking.
- GFRA1 is a glycoprotein with multiple disulfide bonds and a C-terminal GPI-anchor for membrane attachment. Alternative splicing generates several isoforms.
- Mutations in GFRA1 have been associated with disease variants.
- The calculated molecular weight of the mature protein is approximately 51 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 345–420 in the C-terminal region of human GFRA1, suggesting the antibody may recognize the extracellular domain near the GPI anchor site.
- For Western blotting, the expected band is around 51 kDa; however, glycosylation may shift the apparent molecular weight. Validate band specificity with appropriate positive and negative controls.
- In IHC-P, consider antigen retrieval methods and use brain or neuronal tissue sections as positive controls, where GFRA1 is highly expressed.
- For IF/ICC, permeabilization is recommended to detect intracellular pools; co-staining with organelle markers can confirm localization.
- Always titrate the antibody and optimize detection conditions for your specific sample system.
