Biological Background: GET4 Function and Localization

  • GET4 (Golgi to ER traffic protein 4 homolog), also known as conserved edge-expressed protein (CEE) or transmembrane domain recognition complex 35 kDa subunit (TRC35), is a component of the BAG6/BAT3 complex involved in protein quality control.
  • Functions in maintaining misfolded and hydrophobic patch-containing proteins in a soluble state, facilitating their delivery to the endoplasmic reticulum (ER) or targeting them to the proteasome for degradation. Related references: PMID:20676083, PMID:21636303, PMID:21743475
  • Plays a role in the post-translational delivery of tail-anchored/type II transmembrane proteins to the ER membrane by interacting with newly synthesized proteins and cooperating with SGTA and ASNA1. Related references: PMID:20676083, PMID:25535373, PMID:28104892
  • Also serves as a sorting platform for mislocalized secretory pathway proteins, either directing them to the proteasome or to the ER. Related references: PMID:21743475
  • Involved in ER-associated degradation (ERAD), maintaining retrotranslocated proteins in an unfolded soluble state for proteasomal delivery. Related references: PMID:21636303
  • Localizes to the cytoplasm and cytosol.
  • Post-translationally modified by acetylation and phosphorylation, and is subject to alternative splicing; associated with congenital disorder of glycosylation (CDG2Y) based on UniProt keywords.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the full-length human GET4 sequence (amino acids 1-327), suggesting the antibody may detect the target at approximately 37 kDa in Western blotting; consider using whole-cell lysates from human, mouse, or rat tissues/cell lines.
  • For ELISA applications, the antibody may be suitable as a capture or detection reagent; pairing with a matched antibody set can improve assay performance.
  • As a polyclonal antibody, it may recognize multiple epitopes and could potentially cross-react with homologous proteins; validation in your specific experimental system is recommended.
  • Consider optimizing antibody dilution and incubation conditions for each application and sample type.
