Biological Background: GEMIN6 Function and Localization
- GEMIN6 (Gem-associated protein 6), also known as SIP2, is a component of the survival motor neuron (SMN) complex, which is essential for the assembly of small nuclear ribonucleoproteins (snRNPs), the building blocks of the spliceosome.
- The SMN complex, including GEMIN6, catalyzes the assembly of a heptameric Sm protein ring onto small nuclear RNA (snRNA) to form the core snRNP, a critical step in pre-mRNA splicing.
- In the cytoplasm, GEMIN6 participates in the transfer of Sm proteins from the chaperone CLNS1A (pICln) to the SMN complex, facilitating the formation of an intermediate complex that ultimately allows snRNA binding.
- GEMIN6 localizes to the nucleus, nucleoplasm, and specifically to nuclear gems (Gemini of Cajal bodies), as well as the cytoplasm, reflecting its shuttling role in snRNP biogenesis.
- Functionally, GEMIN6 is implicated in mRNA processing and splicing, as highlighted by its association with the spliceosomal machinery and its presence in proteomics datasets.
- The protein is subject to phosphorylation, as indicated by its annotation as a phosphoprotein, which may regulate its activity or interactions within the SMN complex.
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody corresponds to the N-terminal region (amino acids 1–167) of human GEMIN6. Consider that detection may be limited to full-length GEMIN6 or fragments containing this region.
- For western blot (WB) applications, validate the antibody using human-derived cell lysates or tissue extracts known to express GEMIN6, as cross-reactivity with other species is not reported.
- In ELISA, use the recombinant GEMIN6 protein (e.g., the immunogen) as a positive control to confirm antibody specificity and titer.
- Note that GEMIN6 has a predicted molecular weight of ~19 kDa; confirm observed band size in your experimental system, as post-translational modifications may cause slight shifts.
