Biological Background: Gemin 2 Function and Localization
- Gem-associated protein 2 (GEMIN2), also known as SIP1 (Survival of motor neuron protein-interacting protein 1) and SIP1-delta, is a core component of the SMN complex.
- The SMN complex, which includes GEMIN2, catalyzes the assembly of small nuclear ribonucleoproteins (snRNPs), the building blocks of the spliceosome, by accepting Sm proteins from the CLNS1A chaperone and facilitating their assembly onto snRNA, a critical step for pre-mRNA splicing. Related references: PMID:18984161, PMID:9323129
- GEMIN2 constrains the conformation of the 5Sm intermediate, promoting correct snRNA binding and preventing premature assembly, thus ensuring fidelity in snRNP biogenesis. Related references: PMID:16314521, PMID:21816274, PMID:31799625
- Upon snRNA binding, the SMN complex is evicted, allowing the final Sm proteins SNRPD3 and SNRPB to join and complete the core snRNP. Related references: PMID:31799625
- GEMIN2 localizes to both the cytoplasm and the nucleus, specifically accumulating in gems, consistent with its dual role in snRNP biogenesis and splicing.
- The protein undergoes phosphorylation and is subject to alternative splicing and alternative initiation, resulting in multiple isoforms that may contribute to functional diversity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 180–280 of human Gemin 2 (NP_003607.1); this region may be considered for blocking peptide experiments or epitope specificity studies.
- In Western blot, the predicted molecular weight is approximately 32 kDa; however, alternative isoforms or post-translational modifications may produce additional bands.
- The antibody cross-reacts with human, mouse, and rat; users should validate performance in the relevant species and sample type.
- Suitable for applications including WB, IHC-P, and ELISA; optimal dilutions and conditions should be determined empirically.
