Biological Background: GDI1 Function and Localization
- GDI1, or Rab GDP dissociation inhibitor alpha, is also known as Guanosine diphosphate dissociation inhibitor 1 (GDI-1), Oligophrenin-2, and Protein XAP-4.
- It regulates the GDP/GTP exchange reaction of most Rab proteins by inhibiting the dissociation of GDP from them and the subsequent binding of GTP.
- GDI1 promotes the dissociation of GDP-bound Rab proteins from membranes and inhibits their activation; specific Rab targets include RAB1A, RAB3A, RAB5A, and RAB10.
- The protein localizes to the cytoplasm, Golgi apparatus, and trans-Golgi network.
- GDI1 is predominantly expressed in the brain and neural/sensory tissues.
- Mutations in GDI1 are associated with intellectual disability, highlighting its critical role in neuronal function.
- Additional keywords include GTPase activation, direct protein sequencing, proteomics identification, and it is part of the reference proteome.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody is a recombinant fusion protein covering amino acids 188–447 of human GDI1; this region spans the C-terminal portion, which may confer specificity.
- For Western blot (WB), consider using brain tissue lysates or cell lines expressing endogenous GDI1, as the protein is enriched in neural tissues.
- When performing ELISA, ensure optimal coating conditions and validate with appropriate positive and negative controls.
- Cross-reactivity with human and mouse is reported; verify performance in your species of interest if using other models.
