Biological Background: GDF15 Function and Localization
- GDF15, also known as MIC-1, NAG-1, PLAB, PTGFB, and PDF, is a secreted member of the TGF-beta superfamily with hormone and cytokine activities.

Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within the propeptide region (amino acids 100-200); therefore, the antibody is expected to detect the full-length precursor (~34 kDa) on Western blot. The mature secreted form (~12.5 kDa) may not be recognized if the epitope is removed during processing.
- For Western blot, use reducing conditions and load positive control lysates from GDF15-expressing tissues (e.g., placenta, prostate, colon) or cell lines. Optimal antibody dilution should be determined empirically.
- For ELISA, the antibody can be evaluated as a capture or detection reagent; validate pairing with a different anti-GDF15 clone for sandwich assays. Serum or plasma samples are appropriate given the secreted nature of the target.
- Keep samples cold and avoid repeated freeze-thaw cycles to preserve antigen integrity.
