  • GCNT3 (beta-1,3-galactosyl-O-glycosyl-glycoprotein beta-1,6-N-acetylglucosaminyltransferase 3) is a glycosyltransferase also known as C2GnT-mucin type (C2GnT-M) or core 2/core 4 beta-1,6-N-acetylglucosaminyltransferase (C2/4GnT). It plays a central role in O-glycan biosynthesis.
  • Core enzymatic function: mediates core 2 and core 4 O-glycan branching, two critical steps in mucin-type biosynthesis. Additionally, it possesses I-branching enzyme activity, converting linear into branched poly-N-acetyllactosaminoglycans, which introduces the blood group I antigen during embryonic development.
  • Capable of synthesizing all known mucin beta 6 N-acetylglucosaminides, demonstrating broad substrate specificity.
  • Encoded by the GCNT3 gene in humans, mapping to chromosomal region 15q21.3.
  • Subcellular localization: Golgi apparatus membrane, consistent with its role in post-translational glycosylation.
  • Tissue specificity: primarily expressed in mucus-secreting tissues, including colon, kidney, small intestine, trachea, and stomach, where mucin production is essential.
  • Structural features: contains a single-pass transmembrane domain (signal-anchor), disulfide bonds, and is itself a glycoprotein. It belongs to the glycosyltransferase 14 family.
  • Observed molecular weight is approximately 51 kDa; the predicted molecular weight from the amino acid sequence is about 50.9 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 27-180) of human GCNT3, which is predicted to be luminal or extracellular. Consider this when designing detection strategies in various sample types.
  • For Western blot, a specific band at ~51 kDa is expected. Note that post-translational modifications (e.g., glycosylation) may cause shifts; validate band identity in appropriate human tissue lysates such as colon or kidney, or in GCNT3-expressing cell lines.
  • Cross-reactivity is reported for human; for other species, check sequence homology before use.
  • ELISA applications may require optimization of coating antigen and antibody concentrations; consider using recombinant GCNT3 protein or cell lysates as targets.
