Biological Background: GCN2 Function and Localization
- GCN2 (eIF-2-alpha kinase 4) is a metabolic-stress sensing protein kinase that phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (EIF2S1/eIF-2-alpha) in response to low amino acid availability. Related references: PMID:25329545, PMID:32610081
- It serves as an activator of the integrated stress response (ISR), leading to global attenuation of cap-dependent translation while allowing ATF4-mediated reprogramming of amino acid biosynthetic gene expression to alleviate nutrient depletion.
- GCN2 binds uncharged tRNAs and is required for translational induction of PRKCH and for cell cycle arrest via regulation of cyclin D1 and CDKN1A/p21 mRNA translation. Related references: PMID:26102367
- Subcellular localization is primarily cytoplasmic.
- Widely expressed, with notable presence in lung, smooth muscle cells, and macrophages.
- Belongs to the serine/threonine-protein kinase family; contains kinase, histidyl-tRNA synthetase-like, and ribosome binding domains.
- Plays roles in synaptic plasticity, learning, memory formation, neurite outgrowth inhibition, and proapoptotic response to glucose deprivation.
- Exhibits antiviral activity against alphavirus and modulates adaptive immunity to yellow fever virus by promoting dendritic cell autophagy and antigen presentation. Related references: PMID:24310610
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1–300 of human GCN2, a region within the N-terminal domain; this polyclonal antibody may recognize multiple isoforms or post-translationally modified forms.
- For Western blotting (WB), a band at approximately 187 kDa is expected; consider using positive control lysates from amino acid-starved or stress-induced cells to confirm specificity.
- In immunohistochemistry (IHC-P), antigen retrieval optimization and tissue sections from lung or smooth muscle may provide robust staining; validate with appropriate controls.
- For immunofluorescence (IF/ICC) and ELISA, titrate the antibody to determine the optimal working concentration for your specific assay conditions and sample types.
