Biological Background: GCN2 Function and Localization

  • GCN2 (eIF-2-alpha kinase 4) is a metabolic-stress sensing protein kinase that phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (EIF2S1/eIF-2-alpha) in response to low amino acid availability. Related references: PMID:25329545, PMID:32610081
  • It serves as an activator of the integrated stress response (ISR), leading to global attenuation of cap-dependent translation while allowing ATF4-mediated reprogramming of amino acid biosynthetic gene expression to alleviate nutrient depletion.
  • GCN2 binds uncharged tRNAs and is required for translational induction of PRKCH and for cell cycle arrest via regulation of cyclin D1 and CDKN1A/p21 mRNA translation. Related references: PMID:26102367
  • Subcellular localization is primarily cytoplasmic.
