Biological Background: GCN1L1 Function and Localization
- GCN1L1 is a stalled ribosome sensor that plays a key role in the RNF14-RNF25 translation quality control pathway, promoting ubiquitination and degradation of translation factors on stalled ribosomes. Related references: PMID:32610081, PMID:36638793, PMID:37651229
- Following ribosome stalling, GCN1L1 promotes recruitment of RNF14, which directly ubiquitinates EEF1A1/eEF1A, leading to its degradation. Related references: PMID:36638793, PMID:37951215, PMID:37951216
- The RNF14-RNF25 translation quality control pathway, mediated by GCN1L1, also targets ETF1/eRF1 for degradation and ubiquitinates ribosomal protein. Related references: PMID:36638793, PMID:37651229
- GCN1L1 acts as a positive activator of the integrated stress response (ISR) by mediating activation of EIF2AK4/GCN2 in response to amino acid starvation.
- GCN1L1 is a cytoplasmic protein (Subcellular location: Cytoplasm).
- GCN1L1 is ubiquitously expressed, with notable expression in skeletal muscle, ovary, and testis. Related references: PMID:9039502, PMID:9234705
- Post-translational modifications include acetylation and phosphorylation (Keywords: Acetylation, Phosphoprotein).
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to amino acids 2462-2671 of human GCN1L1. Researchers should verify that this epitope is accessible in their experimental system.
- This antibody has been validated for Western blot (WB) and ELISA applications. For other applications, such as IP or IF, it is advisable to perform pilot experiments to confirm suitability.
- Given the large molecular weight of GCN1L1 (293 kDa), ensure appropriate gel percentage and transfer conditions for WB to achieve optimal resolution and transfer efficiency.
