Biological Background: GCN1L1 (GCN1) Function and Localization

  • GCN1L1, also known as GCN1, GCN1-like protein 1, or translational activator GCN1, is a large ~293 kDa protein that functions as a stalled ribosome sensor.
  • It directly binds to ribosomes and acts as a sentinel for colliding ribosomes. Upon ribosome stalling, GCN1 promotes recruitment of the E3 ubiquitin ligase RNF14, which ubiquitinates EEF1A1/eEF1A and other translation factors, targeting them for degradation.
