Biological Background: GCC2 Function and Localization
- GCC2 encodes GRIP and coiled-coil domain-containing protein 2, also known as GCC185, RANBP2L4, and renal carcinoma antigen NY-REN-53.
- It functions as a Golgin that tethers transport vesicles to the trans-Golgi network (TGN) and acts as a RAB9A effector to regulate the recycling of the mannose 6-phosphate receptor from late endosomes to the TGN.
- GCC2 is required for the maintenance of Golgi structure and the biogenesis of noncentrosomal, Golgi-associated microtubules through recruitment of CLASP1 and CLASP2.
- The protein localizes to the cytoplasm, Golgi apparatus, and specifically the trans-Golgi network membrane.
- Ubiquitously expressed across human tissues.
- Post-translational modifications include phosphorylation and acetylation; multiple protein isoforms are generated via alternative splicing.
- With a predicted molecular weight of ~196 kDa (195,910 Da), GCC2 is a large coiled-coil membrane trafficking protein.
