Biological Background: GC1q R/C1QBP Function and Localization
- GC1q R/C1QBP (Complement component 1 Q subcomponent-binding protein, mitochondrial) is a multifunctional protein known as p32, HABP1, gC1qR, and SF2AP32, involved in diverse cellular processes including inflammation, ribosome biogenesis, apoptosis, and splicing. Related references: PMID:10022843 PMID:10479529 PMID:10722602
- It functions as a cell surface receptor for complement C1q (inhibiting C1 activation) and, in complex with cytokeratin-1, serves as a high-affinity receptor for kininogen-1/HMWK. Related references: PMID:10479529 PMID:11859136 PMID:8662673 PMID:21544310
- In mitochondria, it is required for protein synthesis, contributing to assembly of the 55S mitoribosome, and acts as a reader of m5C-modified mitochondrial RNAs to recruit the degradosome under stress. Related references: PMID:28942965 PMID:39019044
- It regulates pre-mRNA splicing by inhibiting the RNA-binding capacity and phosphorylation of SRSF1. Related references: PMID:10022843 PMID:21536856
- GC1q R/C1QBP modulates apoptosis by stabilizing mitochondrial CDKN2A isoform smARF. Related references: PMID:17486078
- It exhibits antimicrobial activity by binding hyaluronan on the cell surface and inhibiting Streptococcus pneumoniae hyaluronate lyase. Related references: PMID:19004836
- The protein localizes to multiple subcellular compartments, including the mitochondrial matrix, nucleus, nucleolus, cell membrane, cytoplasm, and is also secreted.
- It is expressed on the surface of peripheral blood cells, macrophages, and monocyte-derived dendritic cells.
Experimental Guidance and Technical Tips
- The immunogen peptide corresponds to a central region of the human C1QBP protein; cross-reactivity with mouse and rat suggests epitope conservation across these species.
- For Western blotting, expect a band near 31–33 kDa; glycosylated or modified forms may appear slightly larger. Use appropriate controls.
- In IHC-P, optimize antigen retrieval conditions and test on known positive tissues such as kidney, liver, or immune tissues where the target is abundant.
- For ELISA, consider using recombinant C1QBP as antigen; empirical titration of antibody and antigen concentrations is essential.
- Because C1QBP localizes to multiple cellular compartments, subcellular fractionation or co-localization assays may help confirm specificity in your experimental system.
