Biological Background: GBP2 Function and Localization
- GBP2 (Guanylate-binding protein 2), also known as GTP-binding protein 2 (GBP-2) or interferon-induced guanylate-binding protein 2, is a member of the GBP family of GTPases.
- GBP2 functions as an interferon-inducible GTPase that plays important roles in innate immunity against a diverse range of bacterial, viral and protozoan pathogens. Related references: PMID:31091448
- GBP2 hydrolyzes GTP primarily to GDP, with GMP as a minor product, through two consecutive cleavage reactions. Related references: PMID:8706832
- Upon infection, GBP2 is recruited to pathogen-containing vacuoles or vacuole-escaped bacteria, where it promotes vacuole lysis and release of pathogens into the cytosol, thereby positively regulating inflammasome assembly and activation.
- In the cytosol, GBP2 promotes recruitment of proteins that mediate bacterial cytolysis, liberating ligands such as lipopolysaccharide (LPS) and double-stranded DNA (dsDNA) that activate the CASP4/CASP11 and AIM2 inflammasomes, respectively.
- Independently of its GTPase activity, GBP2 acts as an inhibitor of various viruses, including HIV-1, Zika and influenza A, by inhibiting FURIN-mediated maturation of viral envelope proteins. Related references: PMID:31091448
- Subcellularly, GBP2 localizes to the cytoplasmic vesicle membrane, Golgi apparatus membrane, cytoplasm, and perinuclear region.
- GBP2 is prenylated and methylated, and contains a GTP-binding domain, consistent with its role as a membrane-associated GTPase; it is also identified as a lipoprotein.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 410-520 of human GBP2 (NP_004111.2), a region that may provide specific recognition of GBP2; consider validating antibody specificity in the relevant cell or tissue type.
- Given the applications listed (WB, IF/ICC, ELISA), this antibody can be considered for detecting GBP2 in human and mouse samples, but optimal working concentrations should be empirically determined.
- For IF/ICC, ensure appropriate permeabilization and fixation, as GBP2 localizes to cytoplasmic vesicles and perinuclear regions.
- Cross-reactivity with mouse GBP2 suggests potential utility in rodent models; however, confirm sequence homology in the immunogen region to avoid off-target effects.
