Biological Background: GBF1 Function and Localization

  • GBF1 (Golgi-specific brefeldin A-resistance guanine nucleotide exchange factor 1) is a 206 kDa protein encoded by the GBF1 gene (Gene ID: 8729). It belongs to the family of guanine-nucleotide exchange factors (GEFs) for Arf small GTPases.
  • Functions as a GEF for ARF1, ARF4, and ARF5, catalyzing GDP to GTP exchange to activate Arf proteins, which initiates COPI vesicle coating at early secretory compartments. Related references: PMID:12047556, PMID:12808027, PMID:15616190
  • Recruited to cis-Golgi membranes in an Arf-GDP-dependent manner and is essential for COPI coat recruitment at ER exit sites (ERES), ERGIC, and cis-Golgi compartments, mediating retrograde transport to the ER. Related references: PMID:12047556, PMID:12808027, PMID:16926190
  • Involved in trans-Golgi network recruitment of clathrin adaptors (GGA1-3, BIG1/2, AP-1) through GEF activity on ARF4/ARF5, facilitating clathrin-dependent transport. Related references: PMID:23386609
  • Required for Golgi apparatus assembly and maintenance; phosphorylation by AMPK triggers Golgi disassembly during mitosis and stress. Related references: PMID:12808027, PMID:18003980, PMID:18063581, PMID:23418352
  • Subcellularly localized to the Golgi apparatus (cis- and trans-Golgi), ERGIC, cytoplasm, lipid droplets, and membrane.
  • Ubiquitously expressed and linked to Charcot-Marie-Tooth disease type 2GG (CMT2GG) when mutated.
  • May participate in G protein-coupled receptor (GPCR)-mediated chemotaxis and superoxide production in neutrophils by recruiting ARF1 and NADPH oxidase complex to the leading edge. Related references: PMID:22573891
  • Plays a role in maintaining mitochondrial morphology. Related references: PMID:25190516

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-85) of human GBF1, suggesting the antibody may recognize epitopes within this domain.
  • For Western blot, use a high-molecular-weight gel (e.g., 8% SDS-PAGE) and optimize transfer conditions for the large 206 kDa protein.
  • Validate antibody specificity in relevant human and mouse cell/tissue lysates; consider using siRNA knockdown or overexpression controls.
  • For ELISA, the antibody can serve as a capture or detection reagent; titrate to determine optimal working concentration.
