Biological Background: Glycine Amidinotransferase Function and Localization
- GATM (glycine amidinotransferase, also known as AGAT or transamidinase) is a mitochondrial enzyme that catalyzes the transfer of an amidino group from L-arginine to glycine, the rate-limiting step of creatine biosynthesis. Related references: PMID:27233232, PMID:36543883, PMID:3800397
- In addition to glycine, GATM can use beta-alanine, GABA, and taurine as amidino acceptors, producing various guanidine derivatives. Related references: PMID:16820567
- It localizes predominantly to the mitochondrial inner membrane, with some cytoplasmic presence.
- Tissue expression is highest in kidney, but it is also found in brain, heart, liver, lung, salivary gland, and skeletal muscle; biallelic expression occurs in placenta and fetal tissues.
- Alternative splicing generates multiple isoforms, and post-translational modifications include acetylation and phosphorylation; mutations in GATM are associated with human diseases.

- The immunogen spans a large region (aa 38-423), so the antibody may recognize multiple isoforms and species homologs; validate specificity in your target tissue or cell line.
- For Western blotting, the predicted molecular weight of human GATM is ~48 kDa; include appropriate positive and negative controls.
- For immunohistochemistry (IHC-P), antigen retrieval may be required; optimize using known positive tissues such as kidney.
- For immunofluorescence (IF/ICC), mitochondrial localization requires permeabilization; consider using MitoTracker co-staining to confirm.
