Biological Background: GATA6 Function and Localization
-
Transcription factor GATA-6 (GATA-binding factor 6) functions as a transcriptional activator, regulating genes such as SEMA3C and PLXNA2, and is involved in BMP-mediated cardiac-specific gene expression. Related references: PMID:19666519, PMID:22750565, PMID:22824924
-
GATA6 regulates gene expression in the gastric epithelium and may protect epithelial cells from bacterial infection. Related references: PMID:9315713, PMID:16968778
-
In human skin, GATA6 controls sebocyte differentiation, proliferation, and lipid production, mediates retinoic acid effects, and modulates immune and antimicrobial responses. Related references: PMID:33082341
-
GATA6 binds to BMP response element (BMPRE) DNA sequences within cardiac activating regions, contributing to heart development (By similarity).
-
Predominantly localized in the nucleus, consistent with its role as a DNA-binding transcription factor.
-
Contains two C2H2-type zinc-finger domains, facilitating sequence-specific DNA binding and transcriptional regulation; subject to post-translational modifications such as phosphorylation and ubiquitin conjugation.
-
Expressed in heart, gut, and gut-derived tissues; expression is decreased or lost in acne lesions (as reported in skin pilosebaceous units). Related references: PMID:33082341
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 70-160 of human GATA6, a region N-terminal to the first zinc finger. This sequence is relatively conserved between human and mouse, consistent with the observed cross-reactivity. When using this antibody, verify specificity in the relevant species and sample type.
- For Western blot, a band at approximately 60 kDa is predicted; however, post-translational modifications or alternative initiation may result in shifts. Optimize loading and transfer conditions for nuclear proteins.
- In ELISA applications, consider employing the recombinant immunogen as a positive control to assess antibody binding and titer.
- Given GATA6's nuclear localization, use appropriate lysis buffers and sample preparation protocols to ensure efficient extraction of the target protein.
