Biological Background: GATA4 Function and Localization
- GATA4 is a zinc-finger transcription factor that binds the 5'-AGATAG-3' motif and plays a pivotal role in cardiac development and function. It cooperates with TBX5 to bind cardiac super-enhancers, promoting cardiomyocyte gene expression and repressing endocardial/endothelial genes, and participates in BMP-mediated cardiac gene induction. - GATA4 promotes cardiac myocyte enlargement. Related references: PMID:20081228
- GATA4 is required during testicular development. Related references: PMID:21220346
- GATA4 may regulate sphingolipid signaling by controlling sphingosine-1-phosphate lyase expression. Related references: PMID:15734735
- GATA4 localizes to the nucleus, consistent with its role as a transcription factor.
- Mutations in GATA4 are associated with congenital heart diseases including atrial septal defect and cardiomyopathy.
- GATA4 undergoes post-translational modifications such as methylation, which may regulate its transcriptional activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 343-442 of human GATA4, suggesting the antibody may recognize epitopes in this C-terminal region. For Western blot (WB) and other applications, consider using whole cell lysates from human, mouse, or rat cardiac or testicular tissues given the reported cross-reactivity and expression patterns.
- For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), nuclear localization of GATA4 should be observed; perform appropriate fixation and permeabilization. Validate in the relevant sample system based on your experimental context.
- As with any antibody, titrate and optimize working dilutions for your specific assay conditions and sample types.
