- For Western blot, an expected band of approximately 45 kDa may be observed; use a cardiac tissue lysate as a positive control to confirm detection.
- In immunofluorescence and immunohistochemistry (including paraffin sections), nuclear staining consistent with GATA4 localization should be seen; consider using a nuclear counterstain and heat-induced antigen retrieval for IHC-P.
- As a peptide-based polyclonal antibody, cross-adsorption or peptide blocking can be used to validate specificity in ELISA or IF assays.
- The immunogen sequence (aa 341-440, near the C-terminus) is conserved in human, mouse, and rat, supporting the observed cross-reactivity; nonetheless, verify reactivity in the relevant model system.
