Biological Background: Gastrin-releasing peptide Function and Localization

  • Encodes the gastrin-releasing peptide, a secreted neuropeptide that stimulates the release of gastrin and other gastrointestinal hormones (By similarity).
  • Acts as a key mediator of nonhistaminergic itch sensation; contributes to the perception of pruritic stimuli and transmission of itch signals in the spinal cord, and can induce mast cell degranulation.
  • Plays a role in fear memory processing by modulating neuronal activity in the amygdala and auditory cortex; enhances auditory fear memories via GRPR signaling on VIP-expressing cells (By similarity).
  • Also implicated in feeding behavior, respiratory control (sighing), and modulation of neuronal ion channels (By similarity).
  • Undergoes amidation and proteolytic cleavage on pairs of basic residues to generate bioactive peptide forms.
  • Localized to secretory vesicles and secreted; found in neuron projections.
  • The GRP gene undergoes alternative splicing.
  • Predicted molecular weight is approximately 16.2 kDa.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to full-length human GRP precursor (aa 1-148); therefore, the antibody may recognize both the precursor and mature peptide forms. Consider appropriate controls for post-translational processing.
  • For Western blot, expect a band near 16 kDa (predicted size of the precursor). Additional lower molecular weight bands may represent processed forms.
  • For immunohistochemistry and immunofluorescence, validate the antibody in relevant tissue types known to express GRP, such as gastrointestinal tract, spinal cord, and brain regions (amygdala, auditory cortex).
  • For ELISA, use purified recombinant GRP or synthetic peptides as standards; note that the polyclonal nature may necessitate careful optimization of detection conditions.
