- GAP43, also known as neuromodulin, B-50, pp46, is a growth-associated protein primarily expressed in neurons, encoded by the GAP43 gene.
- It is a major component of motile growth cones at the tips of elongating axons and plays a critical role in axonal and dendritic filopodia induction.
- The protein is involved in nerve growth and neural development, contributing to differentiation, growth regulation, and neurogenesis.
- Subcellularly, GAP43 localizes to the cell membrane, growth cone membrane, synapse, filopodium membrane, perikaryon, dendrites, axons, and cytoplasm.
- GAP43 is a lipoprotein modified by palmitoylation and is also a phosphoprotein; it binds calmodulin and is subject to alternative splicing.
- The canonical isoform has a calculated molecular weight of approximately 24.8 kDa, with observed migration at around 25 kDa in SDS-PAGE.
