Biological Background: Gamma-Synuclein Function and Localization
- Gamma-synuclein (SNCG) is a 13331 Da protein belonging to the synuclein family, also known as breast cancer-specific gene 1 protein (BCSG1), persyn, and synoretin. It shares structural homology with alpha- and beta-synuclein and features characteristic repeat motifs.
- Contributes to neurofilament network integrity and may modulate axonal architecture during neuronal development and in adulthood. In vitro, it increases the susceptibility of neurofilament-H to calcium-dependent proteases.
- May also function in modulating the keratin network in skin, suggesting a role beyond the nervous system.
- Activates the MAPK and Elk-1 signal transduction pathway, implicating it in cellular signaling cascades.
- Subcellular localization is predominantly cytoplasmic, with enrichment in the perinuclear region, centrosome, and mitotic spindle.
- Tissue expression is highest in the brain (particularly substantia nigra) and also detected in corpus callosum, heart, skeletal muscle, ovary, testis, colon, and spleen; weak expression in pancreas, kidney, and lung.
- Classified as a phosphoprotein, identified through global proteomics analyses (keywords: Phosphoprotein, Proteomics identification).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide spanning amino acids 50–127 of human gamma-synuclein (NP_003078.2). This central region may enable recognition of both full-length protein and potential cleavage fragments; consider this when interpreting band patterns in Western blot.
- For Western blot (WB), the predicted molecular weight is ~13 kDa, but actual migration may vary due to post-translational modifications. Run appropriate positive controls (e.g., brain tissue lysates) and validate alongside a molecular weight marker.
- For immunohistochemistry on paraffin-embedded tissues (IHC-P), antigen retrieval may be required. Tissues with high expression, such as brain substantia nigra or breast cancer specimens, can serve as positive controls. Optimize antibody dilution and detection system according to your standard protocols.
- In ELISA applications, the antibody can potentially be used for antigen capture or detection in sandwich formats. Pairing with another clone or format is recommended; validate performance in your specific assay matrix.
- Cross-reactivity is documented for human and mouse; when working with other species, cross-reactivity is not guaranteed and should be tested empirically.
