Biological Background: GALP Function and Localization
- GALP, or galanin-like peptide, is a hypothalamic neuropeptide that binds to G-protein-coupled galanin receptors (GALR1, GALR2, GALR3), implicating it in central homeostatic processes, including regulation of gonadotropin-releasing hormone secretion.
- It exhibits potent vasoconstrictor and anti-edema activity in the cutaneous microvasculature, an effect not mediated by GALR1 or GALR2.
- GALP has antimicrobial activity against Gram-negative bacteria, inducing membrane blebbing. Related references: PMID:23537644
- The protein is secreted and undergoes cleavage on pairs of basic residues, with a signal peptide.
- Isoform 2 is specifically expressed in ganglia of ganglioneuroma and ganglioneuroblastoma, and in differentiated tumor cells of neuroblastoma, but not in undifferentiated neuroblasts.
- In the skin, isoform 2 localizes to pericytes covering microvascular arterioles and venules, and in larger vessels to smooth muscle cells; it is absent from endothelial cells.
- Keywords include alternative splicing, antibiotic, antimicrobial, hormone, neuropeptide.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 25–116 of human GALP, encompassing the mature peptide region; this design supports recognition of secreted GALP in physiological samples.
- For Western blotting, the predicted band size is approximately 13 kDa; validate with appropriate positive controls (e.g., tissues expressing GALP) and consider using reducing conditions.
- In ELISA applications, perform titration to optimize antibody and antigen concentrations; cross-reactivity with mouse and rat GALP allows multispecies analysis, but verify in relevant sample systems.
