Biological Background: GALNT3 Function and Localization
- GALNT3 encodes polypeptide N-acetylgalactosaminyltransferase 3 (GalNAc-T3), a type II membrane protein that initiates mucin-type O-glycosylation by transferring N-acetyl-D-galactosamine (GalNAc) to serine or threonine residues of target proteins.
- Catalyzes the first committed step in O-linked oligosaccharide biosynthesis, a modification critical for protein stability, secretion, and cell-surface interactions. Related references: PMID:16638743, PMID:31932717, PMID:8663203
- Substrates include HIV envelope glycoprotein gp120, EA2, MUC2, MUC1A, MUC5AC, fibronectin, and fibroblast growth factor 23 (FGF23), demonstrating diverse roles in viral envelope modification, mucin production, matrix biology, and phosphate metabolism. Related references: PMID:8663203, PMID:9295285, PMID:16638743, PMID:31932717
- Glycosylation of FGF23 by GALNT3 is essential for intact FGF23 secretion; loss-of-function mutations cause hyperphosphatemic familial tumoral calcinosis (HFTC) and hyperostosis-hyperphosphatemia syndrome. Related references: PMID:16638743, PMID:31932717
- Resides in the Golgi stack membrane, consistent with its role in post-translational modification within the secretory pathway.
- Tissue expression is highest in secretory epithelial gland-containing organs—pancreas, skin, kidney, and testis—with lower levels in prostate, ovary, intestine, colon, placenta, and lung.
- Contains a transmembrane helix, a stem region, and tandem catalytic and lectin domains; requires manganese for activity and undergoes alternative splicing and self-glycosylation.
Experimental Guidance and Technical Tips
- The immunogen (aa 43–160) resides in the N-terminal stem/catalytic region; the polyclonal nature likely provides detection across denaturing and native conditions, making it compatible with WB, IHC-P, IF/ICC, and ELISA.
- For Western blot, the predicted molecular weight is ~73 kDa; due to tissue-specific glycosylation, bands may shift. Consider using pancreatic or renal lysates as positive controls and include deglycosylation enzymes to confirm specificity.
- In IHC-P and IF/ICC, anticipate high signal in pancreas, skin, kidney, and testis. Optimize antigen retrieval and blocking for each tissue type, and validate with a relevant negative control.
- For ELISA, use a matched antibody pair recognizing a non-overlapping epitope to avoid competition. Confirm detection of recombinant GALNT3 protein as a standard.
- Although cross-reactive with mouse and rat, verify sequence identity within the immunogen region if interspecies comparisons are planned; run species-matched IgG controls to rule out non-specific binding.
- As a polyclonal preparation, lot-to-lot consistency can vary. For critical quantitative assays, re-validate each new lot with your specific sample system.
