Biological Background: GALNT3 Function and Localization
- GALNT3 (GalNAc-T3) catalyzes the initial step of mucin-type O-glycosylation, transferring N-acetylgalactosamine (GalNAc) from UDP-GalNAc to serine or threonine residues in acceptor proteins. This activity is essential for proper glycoprotein biosynthesis and cellular signaling.
Related references: PMID:8663203, PMID:16638743, PMID:31932717 - It glycosylates diverse substrates, including MUC2, MUC1A, MUC5AC, HIV gp120, fibronectin, and the bone-derived phosphaturic factor FGF23. Glycosylation of FGF23 by GALNT3 prevents its degradation and regulates phosphate homeostasis.
Related references: PMID:8663203, PMID:9295285, PMID:16638743 - The enzyme resides in the Golgi stack membrane, anchored by a single transmembrane helix, ensuring its position in the secretory pathway.
- Expressed predominantly in organs with secretory epithelia—highest in pancreas, skin, kidney, and testis; lower in prostate, ovary, intestine, colon, placenta, and lung.
- Features a lectin domain and requires manganese for catalytic activity; belongs to the glycosyltransferase 2 family.
- Disease-associated variants (HFTC, HHS) underscore its clinical significance in phosphate metabolism and calcification disorders.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 1–140) of human GALNT3, so the antibody may detect both full-length protein and N-terminal isoforms or fragments.
- In Western blot, a band near 73 kDa is expected; however, post-translational modifications or alternative splicing could shift the apparent molecular weight. Validate using a positive control lysate from a tissue with high GALNT3 expression, such as pancreas or kidney.
- For ELISA assays, titrate the antibody and use recombinant GALNT3 protein as a standard. Appropriate blocking and washing conditions should be optimized.
- Since only human reactivity is confirmed, cross-reactivity with other species should be independently tested if needed.
- Keep the antibody at recommended storage conditions and minimize freeze-thaw cycles to preserve activity.
