Biological Background: Galectin 3/LGALS3 Function and Localization
- Galectin-3 (LGALS3), also known as 35 kDa lectin, Mac-2 antigen, CBP35, and L-31, is a member of the galectin family of carbohydrate-binding proteins with specificity for β-galactosides.
- Functions as a pre-mRNA splicing factor in the nucleus and as a galactose-specific lectin in the cytoplasm and extracellular space, binding IgE and laminin.
- Mediates cell–cell and cell–matrix interactions, including integrin-dependent endothelial cell migration stimulated by CSPG4, and cooperates with DMBT1 for terminal differentiation of columnar epithelial cells during embryogenesis.
- Involved in acute inflammatory responses: promotes neutrophil activation and adhesion, chemoattraction of monocytes/macrophages, opsonization of apoptotic neutrophils, and mast cell activation.
- Interacts with TRIM16 to coordinate recognition of endomembrane damage with mobilization of core autophagy regulators ATG16L1 and BECN1.
- Subcellular localization includes cytoplasm, nucleus, and secretion; this distribution is functionally relevant for splicing, intracellular signaling, and extracellular lectin activity.
- Predominantly expressed in colonic epithelium and activated macrophages; also detected in fetal membranes.
- Post-translational modifications include acetylation and phosphorylation, and the protein contains a conserved carbohydrate-recognition domain with a six-stranded β-sandwich fold.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide corresponding to amino acids 100–200 of human Galectin-3 (P17931). Optimize antibody dilution for each application and validate in appropriate positive/negative cell or tissue lysates.
- For WB, expect a band around 26 kDa. Use appropriate loading controls and confirm specificity with knockdown or knockout samples if available.
- In IHC-P and IF/ICC, include proper fixation and antigen retrieval steps; use tissues or cells with known expression (e.g., colon, macrophages).
- Due to cross-reactivity with mouse and rat, this antibody can be used in rodent models after verifying performance.
- For ELISA, use recombinant or purified Galectin-3 standards for quantitation and validate plate-coating conditions.
