- Galectin-3 is a galactose-specific lectin encoded by the LGALS3 gene, also known as MAC2, CBP35, and L31.
- It binds IgE and mediates endothelial cell migration via integrin alpha-3/beta-1, and together with DMBT1, is required for terminal differentiation of columnar epithelial cells during early embryogenesis.
- In the nucleus, galectin-3 acts as a pre-mRNA splicing factor, participating in mRNA processing.
- It is involved in acute inflammatory responses, including neutrophil activation and adhesion, chemoattraction of monocytes/macrophages, opsonization of apoptotic neutrophils, and mast cell activation.
- Galectin-3 coordinates recognition of membrane damage with autophagy by interacting with TRIM16 to recruit ATG16L1 and BECN1 in response to damaged endomembranes.
- Subcellularly, it is found in the cytoplasm, nucleus, and secreted into the extracellular space.
- It is highly expressed in colonic epithelium, activated macrophages, and fetal membranes.
- Post-translational modifications include acetylation and phosphorylation; the protein contains a carbohydrate-recognition domain (CRD) and a repeat structure.
