Biological Background: Galectin-1 Function and Localization
- Galectin-1 (also known as 14 kDa laminin-binding protein, galaptin, HBL, HPL, and S-Lac lectin 1) is a beta-galactoside-binding lectin encoded by the LGALS1 gene.
- It binds a wide array of complex carbohydrates and plays roles in regulating apoptosis, cell proliferation, and cell differentiation.
- Galectin-1 is a strong inducer of T-cell apoptosis and inhibits CD45 protein phosphatase activity, thereby preventing dephosphorylation of Lyn kinase.
- It negatively regulates Th17 cell differentiation via activation of the receptor CD69. Related references: PMID:24752896
- The protein is secreted into the extracellular space and extracellular matrix, and is also found in the cytoplasm.
- It is expressed in a variety of tissues including placenta (trophoblasts, stromal cells), cardiac, smooth, and skeletal muscle, neurons, thymus, kidney, and hematopoietic cells.
- Post-translational modifications include acetylation and phosphorylation, as indicated by UniProt keywords.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human Galectin-1 sequence (amino acids 1–135), which typically yields antibodies recognizing the native protein.
- For Western blotting, a specific band at approximately 15 kDa is expected; validate using appropriate cell or tissue lysates.
- In immunohistochemistry (IHC-P), consider optimizing antigen retrieval protocols for formalin-fixed paraffin-embedded sections.
- ELISA applications may be used for detection or quantification; ensure appropriate blocking and wash conditions.
- The antibody is reported to cross-react with mouse and rat, making it suitable for studies in these species; however, confirm reactivity in your specific experimental system.
