Biological Background: Galectin 1 Function and Localization
- Galectin-1, encoded by the LGALS1 gene, is a 14.7 kDa lectin also known as 14 kDa laminin-binding protein (HLBP14), beta-galactoside-binding lectin L-14-I, Galaptin, and S-Lac lectin 1.
- It binds beta-galactoside and a wide array of complex carbohydrates, playing key roles in regulating apoptosis, cell proliferation, and cell differentiation.
- Galectin-1 inhibits CD45 protein phosphatase activity, thereby preventing the dephosphorylation of Lyn kinase, and acts as a strong inducer of T-cell apoptosis.
- It negatively regulates Th17 cell differentiation through activation of the receptor CD69. Related references: PMID:24752896
- Subcellularly, galectin-1 is found in the cytoplasm and is secreted into the extracellular space and matrix, consistent with its multifaceted signaling roles.
- Tissue expression is broad: placenta (trophoblasts, stromal cells, villous endothelium), cardiac, smooth and skeletal muscle, neurons, thymus, kidney, and hematopoietic cells.
- Post-translational modifications include acetylation and phosphorylation, and the protein forms homodimers, which are critical for its carbohydrate-binding function.
- As a member of the galectin family of beta-galactoside-binding lectins, it participates in numerous biological processes including cell adhesion, immune response, and cancer progression.
Experimental Guidance and Technical Tips
- The immunogen consists of a recombinant protein covering amino acids 1-135 (full-length human galectin-1), so the polyclonal antibody is expected to recognize native and denatured protein in relevant species.
- For Western blotting (WB), validate the antibody in lysates from target tissues or cell lines known to express galectin-1; use appropriate loading controls and consider the predicted molecular weight (approx. 15 kDa).
- In ELISA, this antibody can be used for direct or indirect detection; for sandwich immunoassays, pair with a carefully selected capture antibody after empirical optimization.
- Cross-reactivity with mouse and rat makes this antibody suitable for comparative studies, but confirm performance under your experimental conditions due to potential polyclonal batch variability.
