Biological Background: Galectin-1 Function and Localization

  • Galectin-1 (LGALS1), also known as 14 kDa laminin-binding protein (HLBP14) or S-Lac lectin 1, is a beta-galactoside-binding lectin that belongs to the galectin family.
  • It plays critical roles in regulating apoptosis, cell proliferation, and cell differentiation by binding to beta-galactoside and complex carbohydrates.
  • In immune regulation, galectin-1 inhibits CD45 protein phosphatase activity, sustaining Lyn kinase phosphorylation, and strongly induces T-cell apoptosis. It also negatively regulates Th17 cell differentiation via activation of the receptor CD69. Related references: PMID:24752896
  • The protein is localized to the extracellular space and matrix, as well as the cytoplasm, and is also secreted.
  • Galectin-1 is expressed in a wide range of tissues, including placenta (trophoblasts, stromal cells, endothelium), fetal membranes, cardiac, smooth, and skeletal muscle, neurons, thymus, kidney, and hematopoietic cells.
  • Post-translational modifications include acetylation and phosphorylation, as indicated by proteomics and direct protein sequencing studies.
  • Given its lectin activity and broad tissue expression, galectin-1 may also participate in antimicrobial responses and cell–extracellular matrix interactions.

Experimental Guidance and Technical Tips

  • For Western blot, the predicted molecular weight is approximately 15 kDa. Consider using a 15% SDS-PAGE gel and testing lysates from human, mouse, or rat tissues or cell lines.
  • For immunofluorescence/ICC, include appropriate permeabilization steps since galectin-1 is found both intracellularly and in the extracellular space. Validate fixation and permeabilization conditions in the relevant sample system.
  • When using in ELISA, the antibody can serve as a capture or detection reagent. Note that polyclonal antibodies may show lot-to-lot variation; include appropriate controls and consider cross-reactivity in complex samples.
  • If high background or nonspecific binding is observed, especially in tissue immunohistochemistry, optimize blocking conditions and perform antigen retrieval as needed.
