Biological Background: Galectin-1 Function and Localization
- Galectin-1, encoded by the LGALS1 gene, is a member of the galectin family of carbohydrate-binding proteins, also known as 14 kDa laminin-binding protein or beta-galactoside-binding lectin L-14-I.
- Functions as a lectin that binds beta-galactoside and a wide array of complex carbohydrates, playing key roles in regulating apoptosis, cell proliferation, and cell differentiation.
- Inhibits CD45 protein phosphatase activity, thereby preventing dephosphorylation of Lyn kinase, and acts as a strong inducer of T-cell apoptosis.
- Plays a negative role in Th17 cell differentiation through activation of the receptor CD69. Related references: PMID:24752896
- Subcellular localization includes secretion into the extracellular space and matrix, as well as cytoplasmic localization.
- Post-translational modifications include acetylation and phosphorylation, as identified by direct protein sequencing and proteomics.
- Exhibits broad tissue expression, including placenta, decidua, fetal membranes, cardiac, smooth and skeletal muscle, neurons, thymus, kidney, and hematopoietic cells.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1–100 of human Galectin-1; this region may help ensure recognition of the native protein.
- For Western blot applications, note that Galectin-1 has a predicted molecular weight of ~15 kDa; consider using appropriate gel percentages and validate detection using relevant control lysates.
- When performing immunohistochemistry (IHC-P) or immunofluorescence (IF/ICC), optimal antibody dilution and antigen retrieval conditions should be determined empirically.
- This antibody has cited cross-reactivity with human, mouse, and rat samples; validate species-specific performance in your experimental system.
