Biological Background: GALC Function and Localization
- GALC (galactocerebrosidase), also referred to as galactocerebroside beta-galactosidase or galactosylceramidase, is a lysosomal hydrolase involved in sphingolipid metabolism.
- It hydrolyzes the galactose ester bonds of glycolipids, including galactosylceramide and galactosylsphingosine. Related references: PMID:8281145, PMID:8399327
- GALC is responsible for the lysosomal catabolism of galactosylceramide, a major lipid in myelin, kidney, and epithelial cells of the small intestine and colon. Related references: PMID:8281145, PMID:8399327
- The protein is localized in the lysosome.
- GALC is expressed at the protein level in testis, brain, placenta, kidney, and liver, and is detectable in urine.
- Mutations in GALC cause Krabbe disease (globoid cell leukodystrophy), a severe neurodegenerative lysosomal storage disorder.
- The protein is a glycoprotein with disulfide bonds and alternative splicing; it contains a signal peptide for lysosomal targeting.

- The immunogen is a recombinant fragment encompassing amino acids 40-300 of human GALC, which is expected to generate antibodies targeting the N-terminal region. Due to sequence conservation, cross-reactivity with mouse and rat GALC is plausible.
- For Western blot (WB), the predicted molecular weight of GALC is approximately 77 kDa; a specific band near this size is anticipated.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider optimizing antigen retrieval and antibody dilution. Lysosomal localization may appear as punctate cytoplasmic staining.
- In ELISA applications, this antibody may be utilized as a capture or detection reagent; validate assay conditions in the relevant sample matrix.
