Biological Background: GAD67/GAD1 Function and Localization
- GAD1 encodes glutamate decarboxylase 1, also known as GAD67, a 67 kDa isoform of the GABA-synthesizing enzyme (UniProt Q99259).
- Catalyzes the conversion of glutamate to gamma-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the central nervous system, using pyridoxal 5'-phosphate as a cofactor.
- According to UniProt, some isoforms or variants may be enzymatically inactive as glutamate decarboxylase.
- Highly expressed in the brain, particularly in GABAergic neurons; also present in pancreatic islets, testis, adrenal cortex, and other endocrine tissues.
- Associated with neurological disorders: mutations in GAD1 are linked to epilepsy and intellectual disability (DEE89).
- Undergoes alternative splicing, producing multiple transcript variants; classified as a phosphoprotein and pyridoxal phosphate-dependent decarboxylase.
- Member of the group II decarboxylase family; 3D structure has been determined.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 141-240 of human GAD67/GAD1 (NP_000808.2). For Western blotting, the predicted band is approximately 67 kDa; validate using positive control lysates from brain or pancreatic islet tissues.
- The antibody has been characterized for western blot (WB), immunofluorescence on paraffin-embedded sections (IF-P), and ELISA. Consider appropriate dilutions and blocking conditions based on sample type.
