Biological Background: GAD65/GAD2 Function and Localization

  • GAD65/GAD2 encodes glutamate decarboxylase 2, also known as the 65 kDa glutamic acid decarboxylase (GAD-65).
  • It catalyzes the production of GABA, the major inhibitory neurotransmitter.
  • Subcellularly, it is found in the cytoplasm, cytosol, cytoplasmic vesicles, presynaptic cell membrane, and Golgi apparatus membrane.
  • The protein is a pyridoxal phosphate (PLP)-dependent decarboxylase.
  • It undergoes palmitoylation (lipoprotein) and phosphorylation, which are likely to influence its membrane association and activity.
  • GAD65 is involved in neurotransmitter biosynthesis at synapses.

Experimental Guidance and Technical Tips

  • As the antibody was generated using a recombinant human GAD65/GAD2 immunogen, it is expected to detect both native and denatured GAD65. For Western blotting, a band around 65 kDa should be observed.
  • Given the cytoplasmic and membrane-associated localization of GAD65, appropriate permeabilization and antigen retrieval steps should be optimized for immunofluorescence (IF-P) and immunohistochemistry (IHC-P).
  • Cross-reactivity with mouse and rat has been reported by the supplier; however, validation in the specific tissue or cell system is recommended.
  • For ELISA applications, consider using the antibody as a capture or detection reagent in sandwich ELISA setups, paired with another GAD65-specific antibody.
