Biological Background: GA-binding protein alpha chain Function and Localization
- GA-binding protein alpha chain (GABPA), also known as nuclear respiratory factor 2 subunit alpha (NRF2α) and transcription factor E4TF1-60, is a human transcription factor encoded by the GABPA gene (Gene ID: 2551).
- GABPA binds to purine-rich repeats (GA repeats) and positively regulates transcription of target genes, including the transcriptional repressor RHIT/ZNF205. Related references: PMID:22306510
- During microbial infection, GABPA is necessary for the expression of the Adenovirus E4 gene.
- The protein localizes to the nucleus, consistent with its role in transcriptional regulation.
- Post-translational modification: GABPA is annotated as a phosphoprotein, suggesting regulation by phosphorylation.
- According to the UniProt reviewed entry, the calculated molecular weight of GABPA is 51295 Da (~51.3 kDa), which aligns with the observed 51 kDa band.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide spanning amino acids 50-150 of human GABPA. For Western blotting, consider using nuclear protein extracts and validate the signal at approximately 51 kDa.
- This polyclonal antibody is suitable for ELISA; optimal coating or detection concentrations should be determined empirically.
- Reported cross-reactivity with human and rat; performance in other species must be verified.
- As GABPA is a DNA-binding transcription factor, avoid excessive sample heating and consider adding reducing agents during sample preparation to maintain solubility.
