Biological Background: GABARAP Function and Localization
- GABARAP (Gamma-aminobutyric acid receptor-associated protein), also known as MM46 or ATG8A, is a ubiquitin-like modifier belonging to the ATG8 family.
- It plays a critical role in the intracellular transport of GABA(A) receptors and their interaction with the cytoskeleton. Related references: PMID:9892355
- GABARAP is essential for a later stage in autophagosome maturation, distinguishing its function from LC3 proteins that act in phagophore elongation. Related references: PMID:15169837, PMID:20562859, PMID:22948227
- It is required for the local activation of the CUL3(KBTBD6/7) E3 ubiquitin ligase complex, regulating TIAM1 degradation and downstream RAC1 signaling, thereby influencing cell migration and proliferation. Related references: PMID:25684205
- GABARAP participates in selective autophagy of the endoplasmic reticulum (reticulophagy/ER-phagy) through interactions with TEX264 and UBAC2, and has been implicated in apoptosis. Related references: PMID:31006538, PMID:15977068, PMID:39284914
- Subcellularly, GABARAP localizes to autophagosome membranes, cytoplasmic vesicles, the Golgi apparatus membrane, and the cytoskeleton.
- Tissue expression is broad, with high levels in heart, brain, placenta, liver, skeletal muscle, kidney, and pancreas.
- GABARAP is a lipidated ubiquitin-like modifier, anchoring to autophagosomal membranes to execute its functions.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide corresponding to amino acids 1-100 of human GABARAP. For Western blot (WB), consider using cell or tissue lysates known to express GABARAP, such as brain or heart samples, with the predicted band at ~14 kDa. Validate in the relevant sample system to confirm specificity.
- For immunohistochemistry (IHC-P), optimize antigen retrieval conditions and validate in appropriate positive and negative control tissues, as GABARAP is broadly expressed.
- For ELISA, consider pairing with a capture antibody if used in sandwich format, and test with recombinant GABARAP protein or cell lysates.
- Given the high homology among GABARAP family members, cross-reactivity with GABARAPL1 or GABARAPL2 should be evaluated if specificity is critical.
- The antibody is provided as rabbit polyclonal; store at -20°C upon receipt and avoid repeated freeze-thaw cycles.
