Biological Background: GABARAP Function and Localization
- GABARAP (Gamma-aminobutyric acid receptor-associated protein, also known as MM46, ATG8A) is a ubiquitin-like modifier involved in intracellular transport and autophagy.
- It mediates the trafficking of GABA(A) receptors and their interaction with the cytoskeleton. Related references: PMID:9892355
- In autophagy, the GABARAP/GATE-16 subfamily is essential for the maturation stage of autophagosomes, distinguishing its role from the LC3 subfamily. Related references: PMID:15169837, PMID:20562859, PMID:22948227
- GABARAP participates in selective ER-phagy by interacting with reticulophagy receptors TEX264 and UBAC2 to facilitate endoplasmic reticulum turnover. Related references: PMID:31006538, PMID:39284914
- It is required for the activation of the CUL3(KBTBD6/7) ubiquitin ligase complex that targets TIAM1 for degradation, thereby regulating RAC1 signaling and downstream effects on cytoskeleton, cell migration, and proliferation. Related references: PMID:25684205
- GABARAP is also implicated in apoptosis. Related references: PMID:15977068
- It is localized to autophagosome and Golgi membranes, cytoplasmic vesicles, and the cytoskeleton, and is broadly expressed in tissues including brain, heart, liver, and kidney.
Experimental Guidance and Technical Tips
- The immunogen lies within the N-terminal 1-100 amino acid region of human GABARAP (O95166). For Western blot experiments, consider that the monomeric protein has a predicted molecular weight of 14 kDa, but post-translational lipidation may alter its migration pattern; validate the observed band in your sample system.
- When performing ELISA, use the immunogen peptide or recombinant GABARAP as a positive control to confirm antibody binding specificity.
- This antibody is stated to cross-react with mouse and rat; however, it is advisable to verify reactivity in your specific experimental system and tissue type before large-scale studies.
