Biological Background: GAB1 Function and Localization
- GAB1 (GRB2-associated-binding protein 1) is an adaptor protein also known as GRB2-associated binder 1 or Growth factor receptor bound protein 2-associated protein 1.
- It transduces signals from activated receptor tyrosine kinases such as FGFR1, EGFR, and the insulin receptor (INSR).
- GAB1 is critically involved in the MET/HGF-signaling pathway. Related references: PMID:29408807
- Alternative splicing of GAB1 yields multiple isoforms, and the protein is regulated by post-translational modifications including acetylation and phosphorylation.
- Mutations in GAB1 are linked to autosomal recessive non-syndromic deafness (DFNB26).
- The calculated molecular mass of GAB1 is 76.6 kDa, and it migrates at 76–80 kDa in SDS-PAGE.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C‑terminal region (aa 594–694) of human GAB1; this region may contain conserved epitopes explaining the observed mouse cross‑reactivity.
- For western blot, consider a starting dilution of 1:500–1:2000 and validate in a lysate known to express the target.
- ELISA can be optimized for detection of recombinant or endogenous GAB1; pilot experiments with different coating conditions are recommended.
- Confirm the detected band matches the expected 76–80 kDa range, and include positive/negative controls to verify specificity.
- Multiple isoforms exist due to alternative splicing; the antibody may recognize several if the epitope is shared.
