Biological Background: G3BP1 Function and Localization
- G3BP1 (Ras GTPase-activating protein-binding protein 1), also known as GAP SH3 domain-binding protein 1 (HDH-VIII), is a multifunctional protein involved in stress granule formation, innate immunity, and nucleic acid unwinding.
- Plays an essential role in stress granule formation by promoting the formation of membraneless organelles through RNA-binding-induced liquid-liquid phase separation (LLPS). - Functions as an ATP- and magnesium-dependent DNA/RNA helicase (hDH VIII) that unwinds DNA/DNA, RNA/DNA, and RNA/RNA duplexes with 5'→3' directionality, preferentially on substrates with 3' overhangs. Related references: PMID:9889278
- Participates in innate immune signaling: promotes the DNA binding and condensation of cGAS to activate the cGAS/STING pathway, and enhances RIG-I-mediated type I interferon production upon sensing pathogenic RNA. Related references: PMID:30510222, PMID:30804210
