Biological Background: Fyn Function and Localization
- Fyn is a non-receptor tyrosine-protein kinase belonging to the Src family, also known as proto-oncogene Syn, c-Fyn, SLK, and p59-Fyn. Two main isoforms exist: isoform 1 is highly expressed in the brain, while isoform 2 is predominantly found in hemopoietic lineages, especially T-lymphocytes.
- It plays a central role in regulating cell growth, survival, adhesion, integrin-mediated signaling, cytoskeletal remodeling, cell motility, immune response, and axon guidance. Related references: PMID:11536198, PMID:15489916, PMID:15557120
- Fyn is activated by dephosphorylation of its C-terminal inhibitory tyrosine and subsequent PKA-mediated phosphorylation, leading to association with PTK2/FAK1 and targeting to focal adhesions. Related references: PMID:15489916

Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human Fyn, which may allow detection of both major isoforms (isoform 1 and isoform 2). Verify isoform specificity in your experimental system.
- For western blotting (WB), a band around 61 kDa is expected. However, post-translational modifications and isoform expression patterns may affect migration; validate in relevant sample lysates.
- In ELISA applications, consider using this antibody as a capture or detection reagent after confirming performance with your target antigen and buffer conditions.
- Cross-reactivity with mouse and rat has been reported. If working with these species, confirm epitope conservation and run appropriate controls to avoid non-specific binding.
