Biological Background: FXYD3 Function and Localization
- FXYD3 is a FXYD domain-containing protein, also designated mammary tumor 8 kDa protein (Mat-8) or phospholemman-like.
- It associates with and modulates the sodium/potassium-transporting ATPase (NKA) which transports Na(+) out of the cell and K(+) into the cell. Related references: PMID:17077088
- It reduces glutathionylation of the NKA beta-1 subunit ATP1B1, thereby reversing inhibition of the pump. Related references: PMID:21454534
- When expressed in Xenopus oocytes, it induces a hyperpolarization-activated chloride current. Related references: PMID:7836447
- FXYD3 alters the apparent ion affinities of NKA: it decreases K+ affinity over a range of membrane potentials and increases Na+ affinity at slightly negative and positive membrane potentials.
- The protein localizes to the cell membrane and undergoes alternative splicing, yielding isoform 1 (predominantly in differentiated cells) and isoform 2 (mainly in undifferentiated cells).
- Proteomics identification and direct protein sequencing confirm its transmembrane topology and signal peptide.
